Apportunistic identification of a free-living Acanthamoeba spp. clinical in different animales and human in Karbala and Qadisiyah province / Iraq

Abstract

This study was performed in Karbala and Qadisiyah governarat middle of Iraq in perioed from Septemper to October 22 order to detection of the opportunistic Free living amoebae (FLA) and identified by morphological characters in  culture, Polymeras chain reaction (PCR) by specific primers .123 samples were collected from clinical  cases including, eye, skin, nose from animals and human, and CSF  from human .All samples were cultured in the  Non Nutrient agar  medium (NN-agar medina).and then examined by light microscope to recognized Morphologically the trophozoite and cyst of opportunistic amoebas over all 50/123 (43.6%) that belong the genera Acanthamoeba  spp. (A. triangularis, A .astronyxis, A. Polyphaga, A. Castellani). 22/22 (100%) Positive to specific primer to Acanthamoeba spp.  by (PCR) .


Over all 24/53 (19.5%) Postive from eye sampels ,13/36 (10.5%) from skin, 12/26 ( 9.7%)  from nose of clinical animals and human with 1/8 (1%) CSF of human confirmed by PCR   Acanthamoeba spp.  Single-celled living organism that can cause severe infections of the eye , skin ,CNS. and cause fatal encephalitis and blinding  keratitis.
